Pharmacology
Triptorelin is a Gonadorelin analogue, also known as Leuteinzing hormone releasing analogue (GnRH analogue, LHRH analogue). The drug binds to receptors in the pituitary gland and stimulates secretion of gonadotrophins (namely luteinzing hormone and Follicle-stimulating hormone). This causes an initial phase of LH and FSH stimulation, prior to down-regulation of the gonadotrophin-releasing hormone receptors, thereby reducing the release of gonadotrophins in the long term, which in turn leads to the inhibition of androgen and oestrogen production.
Mechanism
Triptorelin is a synthetic decapeptide agonist analog of luteinizing hormone releasing hormone (LHRH). Possessing greater potency than endogenous LHRH,Triptorelin reversibly represses gonadotropin secretion. After chronic, continuous administration, this agent effects sustained decreases in LH and FSH production and testicular and ovarian steroidogenesis. Serum testosterone concentrations may fall to levels typically observed in surgically castrated men. Triptorelin is indicated for the palliative treatment of advanced prostate cancer. It has a role as
a gonadotropin releasing hormone agonist, an antineoplastic agent and a contraceptive drug.
